Mad science and genetic splicing are a consistent theme in the Fallout Universe. Deathclaws were genetically modified Jackson's Chameleons, and New Vegas featured Coyote-Rattlesnake crossbreeds. Bringing back and Mammoth and mutating it to be several times larger doesn't strike me as much of a stretch.
